E-cadherin is a transmembrane protein important for epithelial cell adhesion. It maintains the structural and functional integrity of epithelial tissues. Reduced or aberrant E-cadherin expression disrupts cell-cell contacts, allowing cells to migrate. In normal tissues, E-cadherin is localized to the membrane of epithelial cells, consistent with its role in cell adhesion. In tumor tissues, E-cadherin stains positively in glandular epithelium and in adenocarcinomas of the lung, gastrointestinal tract, and ovary. It has also been detected positively in some thyroid carcinomas. In combination with p120 or cytokeratins, it is involved in the differentiation of ductal (E-cadherin-positive) and lobular (E-cadherin-negative) breast carcinomas. Several studies have reported that reduced E-cadherin expression in various carcinomas correlates with poor prognosis.
