Programmed death 1 (PD-1; CD279) is a transmembrane protein that acts as a surface receptor for its ligands PD-L1 and PD-L2. PD-1 is primarily expressed on activated T cells, B cells, and myeloid cells. PD-1 is an immune checkpoint and plays a crucial role in downregulating the immune system by inhibiting T cell activation, which in turn reduces autoimmunity. Some tumor cells utilize this mechanism to prevent apoptosis by inhibiting the T cells that target them. PD-1 is a useful marker for angioimmunoblastic lymphomas. Furthermore, increased PD-1 expression has been found to be associated with a poor prognosis in hepatocellular carcinoma and renal cell carcinoma. Treatments targeting PD-1 have shown encouraging results in breast cancer, non-small cell lung cancer (NSCLC), and melanoma.
