SATB2 (Special AT-rich sequence-binding protein 2) is a nuclear matrix protein that influences craniofacial formation mechanisms, such as the development of the jaw and palate, and is part of a transcriptional network that regulates skeletal development and osteoblast differentiation. It is highly expressed in the adult and fetal brain and weakly expressed in the liver, kidney, and spinal cord, as well as in some brain regions. SATB2 has been shown to identify colorectal carcinomas, including poorly differentiated colorectal carcinomas and metastases. Adenocarcinomas such as breast, lung, and ovary may express SATB2 at lower frequencies. Therefore, SATB2 is useful for identifying a carcinoma of colorectal origin when a cancer of unknown origin is identified.
